How to read this note

1. Workflow structure
This workflow separates FFPE sample pretreatment from the downstream magnetic RNA purification steps. It is intended as a practical companion to the product manual rather than a replacement for the official protocol. The downstream steps follow a magnetic bead-based RNA purification workflow with DNase I treatment integrated after the first magnetic wash stage.

2. Time interpretation
Protocol times stated in the product manual are retained where applicable. Steps without explicit timing are estimated for an experienced operator. For short protocol ranges, the timeline uses the midpoint. The workflow time shown here follows the manual or liquid-station protocol rather than the automated plate program.

3. Workflow characteristics
This workflow combines short FFPE RNA pretreatment, magnetic bead binding and DNase I treatment in a single RNA-only route. Compared with column-based FFPE RNA workflows, the downstream purification relies on magnetic separation rather than membrane loading, and the DNase step is integrated while RNA remains associated with magnetic particles.

4. Practical considerations
RNA recovery from FFPE material is strongly affected by fixation history, storage condition and heat exposure during pretreatment. The 80°C step should not be extended beyond the intended workflow because prolonged incubation may increase RNA degradation. During magnetic purification, complete liquid removal after washes and controlled bead drying are important for reducing ethanol carryover before elution.
